25h time lapse movie of a developing zebrafish retina, showing the lineage of multiple H2B-GFP cells transplanted into a wild type embryo. The movie started at 24 hours post fertilisation, when all the cells in the retina are proliferating progenitor cells. By identifying single H2B-GFP labelled cells and follow their lineage over time, the researchers are able to track the lineage of these single cells (which cell types does this progenitor give rise to), but also to determine other parameters such as how many times the cell divides, the cell cycle time, migration patterns, etc. This sort of analysis requires the ability to perform fast, long term imaging (24 to 28hpf), with good resolution and less photo-bleaching.
